Descripción General
Oxitocina (OXT) es una hormona nonapéptida cíclica y neuropéptido con la secuencia Cys-Tyr-Ile-Gln-Asn-Cys-Pro-Leu-Gly-NH₂, que presenta un puente disulfuro entre Cys¹ y Cys⁶ y una amida C-terminal (PM 1007,19 Da).[1]
Importancia histórica: Las propiedades contráctiles uterinas de los extractos hipofisarios fueron descubiertas por Sir Henry Dale en 1906. En 1953, Vincent du Vigneaud secuenció y sintetizó la oxitocina — la primera hormona polipeptídica jamás secuenciada o sintetizada — obteniendo el Premio Nobel de Química (1955).[2]
La oxitocina se sintetiza en neuronas magnocelulares y parvocelulares de los núcleos paraventricular (PVN) y supraóptico (SON) del hipotálamo, derivada del gen OXT (cromosoma 20) como una preprooxitocina inactiva que incluye la proteína transportadora neurofisina I, y almacenada en la hipófisis posterior.[3]
Estructuralmente, la oxitocina difiere de la arginina vasopresina (AVP) en solo 2 aminoácidos (Ile³/Leu⁸ en OXT vs Phe³/Arg⁸ en AVP), lo que genera una reactividad cruzada significativa en los receptores de vasopresina.[3]
